急性灰白髄炎（きゅうせいかいはくずいえん、poliomyelitis）は、ポリオ (Polio) とも呼ばれる、ピコルナウイルス科、エンテロウイルス属のポリオウイルスによるウイルス性感染症。ポリオは、Poliomyelitis（ポリオマイアライティス）の省略形。ポリオウイルスが原因で、脊髄の灰白質（特に脊髄の前角）が炎症を起こす。症例の約75%は無症候性である。 発生する可能性のある軽度の症状には、喉の痛みや発熱が含まれる。 症例の割合で、頭痛、首のこわばり、感覚異常などのより重篤な症状が発症する。これらの症状は通常1〜2週間以内に回復する。一般的でない症状は、永久的な麻痺であり、極端な場合には死亡する可能性がある。 回復から数年後、ポリオ後症候群が発生する可能性があり、最初の感染時に人が持っていたものと同様の筋肉衰弱の発達が遅い。初めの数日間は胃腸炎のような症状が現れ、その後1パーセント以下の確率で、ウイルスに関連した左右非対称性の弛緩性麻痺（下肢に多い）を呈する病気である。
