遺伝子発現の調節(いでんしはつげんのちょうせつ)には、細胞が特定の遺伝子産物(タンパク質やRNA)の合成を増加または減少させる幅広いメカニズムが含まれる。生物学において、遺伝子発現のための高度なプログラムは、発達経路の誘導、環境刺激への応答、新たな食料源への適応など、幅広い現象で観察される。転写の開始からRNAのプロセシング、そしてタンパク質の翻訳後修飾に至るまで、遺伝子発現のあらゆるステップが事実上調節が可能である。遺伝子発現の制御因子はしばしば相互に影響し合い、遺伝子調節ネットワークを形成している。

活性化されたIRS-1は、インスリンによって調節される遺伝子の転写を促進するための、細胞内のセカンドメッセンジャーとして機能する。まず、Grb2(英語版)タンパク質のSH2ドメインがIRS-1のリン酸化チロシン残基に結合する。Grb2はSOS(英語版)に結合できるようになり、SOSはGタンパク質であるRasに結合しているGDPのGTPへの交換を触媒する。これによって活性化されたRasはリン酸化カスケードを開始し、最終的に活性化されたMAPKは核へ移行して核内のさまざまな転写因子 (Elk1(英語版)など) をリン酸化する。

「ATP感受性カリウムチャネル」の記事における「遺伝子発現の調節」の解説
KATP遺伝子ファミリーのメンバーとして、4つの遺伝子が同定されている。ABCC8(SUR1)とKCNJ11(Kir6.2)遺伝子は11番染色体(英語版)p15.1に位置し、ABCC9(SUR2A、B)とKCNJ8(Kir6.1)遺伝子は12番染色体(英語版)p12.1に位置する。SUR2AとSUR2Bは選択的スプライシングによる産物である。 これらの遺伝子の転写の変化、すなわちKATPチャネルの産生の変化は、代謝環境の変化と直接関係している。例えば、グルコース濃度が高い時にはKCNJ11のmRNAレベルの大幅な低下が誘導され、グルコース濃度が低い時にはその逆となる。同じように、ラットの左心室の心筋では60分の虚血後24時間から72時の再灌流によってKir6.2の遺伝子の転写が増大する。 低酸素や虚血に対する細胞のKATPの応答に関しては次のような機構が提唱されている。細胞内の酸素レベルの低下は、ミトコンドリアのTCA回路を減速させることで代謝速度を低下させる。効率的に電子伝達を行うことができないため、細胞内のNAD+/NADH比は低下し、PI3キナーゼや細胞外シグナル調節キナーゼが活性化される。その結果、c-Jun(英語版)の転写がアップレギュレーションされ、SUR2の遺伝子のプロモーターに結合するタンパク質が合成される。 細胞の酸化ストレスとKATPの産生の増加との関係が持つ重要な意味の1つは、全体的なカリウム輸送機能がこれらのチャネルの膜濃度と正比例するという点である。糖尿病の場合、KATPチャネルが正常に機能しないため、細胞は不利な酸化的条件に適応できず、軽度の心筋虚血や低酸素に対して顕著に感受性となる。

p300とCBPは3つの方法で遺伝子発現の増加をもたらすと考えられている。 ヒストンアセチルトランスフェラーゼ(HAT)活性による遺伝子プロモーターのクロマチン構造の緩和 プロモーターへのRNAポリメラーゼIIを含む基本転写装置のリクルート アダプター分子としての作用 p300は転写因子に直接結合することで転写を調節する。この相互作用はp300の1つ以上のドメインによって媒介される。p300のKIX、TAZ1、TAZ2、IBiDはそれぞれ、転写因子p53の2つの9aaTADにまたがる配列に強固に結合する。 p300とCBPは遺伝子の転写を調節するエンハンサー領域にも結合することが知られており、これらのタンパク質を用いたChIP-seq(英語版)はエンハンサー領域の予測に利用される。 p300が結合する領域の70%は、DNase I高感受性領域(英語版)との関係が観察されるように、開いたクロマチン領域であることが示されている。さらに、結合部位の75%は転写開始部位から離れており、これらの結合部位はH3K4me1(英語版)に富むことから観察されるようにエンハンサー領域と関係している。RNAポリメラーゼIIのエンハンサー領域での結合部位とp300の結合部位にはある程度の相関があり、この相関はプロモーターとの物理的相互作用またはエンハンサーRNA(英語版)によって説明される可能性がある。
