SNAP25(Synaptosomal-associated protein, 25kDa)は、ヒトではSNAP25遺伝子にコードされるt-SNAREタンパク質である[5]。SNAP25はトランスSNARE複合体の構成要素であり、シナプス小胞と細胞膜との膜融合の特異性を担うとともに、緊密な複合体を形成することで両者の融合を実行する[6]。

構造と機能

神経伝達物質の放出時にエキソサイトーシスを駆動する分子装置。コアSNARE複合体は、シナプトブレビン、シンタキシン、SNAP-25からの4本のαヘリックスによって形成される。シナプトタグミンはCa2+センサーとして機能し、SNAREのジッピング(zipping)を調節する[7]。

Q-SNAREタンパク質であるSNAP-25は、分子中央部のシステイン残基に共有結合したパルミトイル側鎖を介して細胞膜の細胞質側に固定されている。このことは、SNAP25は膜貫通ドメインを持たないことを意味する[8]。

SNAP-25は、4本のαヘリックスドメインからなるSNARE複合体に2本のαヘリックスを提供している因子として同定された[9]。SNARE複合体は小胞融合に関与し、細胞膜への小胞のドッキングと融合によってエキソサイトーシスが行われる。小胞関連膜タンパク質(英語版)(VAMP)ファミリーのシナプトブレビン(英語版)、そしてシンタキシン1(英語版)もそれぞれ1本ずつαヘリックスを提供することでSNARE複合体の形成を助ける。SNAP-25はシナプトブレビン、シンタキシン1とともに組み立てられ、これらの選択的な結合により正確な位置での小胞のドッキングと融合が行われる[10]。

SNARE複合体の形成の際には、シナプトブレビン、シンタキシン1、SNAP-25は結合し、互いに巻き付いてコイルドコイル構造を形成し始める。シナプトブレビンとシンタキシン1の双方のαヘリックスがSNAP25のαヘリックスと結合する。シナプトブレビンはSNAP-25のαヘリックスのC末端近傍に結合するが、シンタキシン1はN末端近傍に結合する[8]。

SNAP-25はシナプス前のP型(英語版)、Q型(英語版)、L型(英語版)の電位依存性カルシウムチャネルを阻害し[11]、シナプトタグミン(英語版)のC2BドメインにCa2+非依存的に結合する[12]。SNAP-25はグルタミン酸作動性シナプスではCa2+応答性を低下させるが、GABA作動性シナプスには存在しない[13]。

SNAP-25には2つのアイソフォーム(mRNAのスプライスバリアント)が存在し、それぞれSNAP25A、Bと呼ばれている。2つのアイソフォーム間には9アミノ酸残基の差異が存在し、4つのシステイン残基のうちの1つも位置が異なる[14]。両者の主要な特徴は下の表に記されている。

臨床的意義

マウスのSNAP-25の遺伝子のヘテロ接合型欠失によって、注意欠陥・多動性障害(ADHD)に類似した活動過多の表現型が生じる。このことは、SNAP-25がシナプスでのCa2+応答性を調節していることと一致する。ヘテロ接合型マウスでは、ADHD治療薬アデロール(英語版)の有効成分であるデキストロアンフェタミン(英語版)(デキセドリン)によって活動過多の緩和が観察される。SNAP-25のホモ接合型欠失は致死である。その後の研究によって、ヒトのSNAP25遺伝子の変異の少なくとも一部はADHDの素因となる可能性が示唆されている[16][17]。

ゲノムワイド関連解析によって、この遺伝子のrs362584多型が神経症傾向と関係している可能性が指摘されている[18]。

ボツリヌストキシンA、C、EはSNAP-25を切断し[19]、ボツリヌス症の麻痺を引き起こす。
