ソルターゼ

ソルターゼ (Sortase) は、原核生物の酵素の一群の一つである。細胞表面タンパク質の識別シグナルのC末端を認識してこれを切断し、表面タンパク質を改変する。ソルターゼの基質の多くは、第一にLPXTGモチーフ(Leu-Pro-any-Thr-Gly)で構成された識別シグナルであり、次いで高度に疎水的な膜貫通タンパク質配列、アルギニンといった塩基性残基のクラスターである。切断はトレオニン残基とグリシン残基の間で起こり、ソルターゼの活性部位のシステイン残基を標的のトレオニン残基に一時的に結合させ、ソルターゼを細胞壁成分に共有結合させる。ソルターゼは全てのグラム陰性細菌と一部のグラム陽性細菌(例えばShewanella putrefaciens)と一部の古細菌(例えばMethanobacterium thermoautotrophicum)で確認されている[1][2] 。ハウスキーピング酵素のソルターゼAは多くの標的タンパク質で典型的なソルターゼの活性を示すが、その他のものは切断モチーフの変異体を認識し、あるいはピリンの繊毛への組み立てを触媒する[3][4][5]。
