AD-1211は、住友ファーマが1970年代に開発したオピオイド鎮痛薬である。化学的には1-置換-4-プレニル-ピペラジン誘導体であり、構造的には他のほとんどのオピオイド薬とは無関係である。このシリーズの(S)-エナンチオマーはオピオイドアゴニストとしてより作用が強いが、この化合物の作用が弱い(R)-エナンチオマーであるAD-1211は、オピオイド受容体において混合アゴニスト・アンタゴニストとして作用し、ペンタゾシンと同様の薬理学的特性を示す[1]。さらに動物実験において、長期投与後も耐性や依存症の発現がほとんどない非定型オピオイド作用を持つ[2][3]。
