RETはGDNFファミリーの細胞外シグナル伝達分子を結合する受容体型チロシンキナーゼであり、ヒトではRET遺伝子にコードされる[5]。RET遺伝子の機能喪失型変異はヒルシュスプルング病の発症と関係しており[6]、機能獲得型変異は甲状腺髄様癌、多発性内分泌腺腫症2A型と2B型を含む、さまざまなタイプのがんの発症と関係している[7]。
