SUMO1(small ubiquitin-related modifier 1)は、ヒトではSUMO1遺伝子によってコードされているタンパク質である[5][6]。
機能
SUMO1遺伝子にコードされるSUMO1は、SUMOタンパク質ファミリーの一員である。ユビキチン様タンパク質の1つであり、翻訳後修飾系の一部として標的タンパク質に結合するという点で、ユビキチンと類似した形で機能するタンパク質である。しかしながら、ユビキチンが主にタンパク質のプロテアソーム分解への標的化と関連しているのとは異なり、SUMO1は核輸送、転写調節、アポトーシス、タンパク質の安定性など広範囲にわたる細胞過程に関与している。SUMO1はタンパク質のC末端の4アミノ酸が切除されることで活性化される。また、SUMO1遺伝子の偽遺伝子がいくつか報告されている[7]。
口唇口蓋裂と関連した遺伝子の多くが、SUMO化経路の構成要素をコードしている[8]。患者の染色体異常の解析により、SUMO1は口唇口蓋裂関連遺伝子座として同定されている[9]。
心臓における役割
心不全は、心臓のポンプ機能が大きく低下し、体が十分な血液循環を行うことができなくなっている状態である。心機能の低下は、疲労、運動耐容能の低下、息切れといった症状を引き起こす。心不全を抱える患者は正常な心機能を有する人と比較して死のリスクが有意に高まる。心不全は世界的に発生率が上昇傾向にあることから公衆衛生上の重大な懸念の1つとなっており、また先進国における主要な死因の1つである[10]。
SUMO1は心機能における重要な構成要素の1つであり、心筋細胞のミトコンドリアにおけるカルシウム恒常性の調節を補助している。SUMO1は、心機能に必要不可欠なタンパク質の1つであるSERCA2aと呼ばれるタンパク質に結合する。SERCAは心筋細胞の筋小胞体に位置する膜貫通タンパク質であり、主な機能は細胞質基質と筋小胞体内腔の間でなされるカルシウムの取り込みと放出の調節である。カルシウムは心筋細胞の収縮と弛緩に必要不可欠な因子であり、SERCA2aによる細胞内のカルシウム恒常性の管理は心機能全般に重要である[11]。通常、SUMO1はSERCA2aのリジン480番と585番に結合してSERCA2aの活性化と安定化を担っており、SUMO1とSERCA2aとの相互作用は心筋細胞内のカルシウム濃度の調節に重要な役割を果たしている。SUMO1タンパク質の減少はSERCA2aの減少をもたらし、それによって心不全を抱える患者ではカルシウムの調節効率が低下する[12]。
創薬標的として
SUMO1は、心不全を抱える患者の心機能の改善のための重要な創薬標的となる可能性がある。マウスモデルでは、遺伝子治療によるSUMO1の導入はSERCA2a活性の改善と関連し、心収縮の増大を介して心機能の改善をもたらすことが報告されている[12]。さらに、SUMO1の過剰発現によってカルシウムの取り込みが加速されることも、心臓細胞内での十分なカルシウム濃度の維持における重要性を示す証拠となっている[12]。
